I assume you're talking about playback in Nero MediaHome.
Is v14 up to date as determined by running the check for updates from the Nero ControlCenter? -

Yes Nero MediaHome 16 and up to date. The mkv file is an x-matroska in the type of file listing on MediaHome.

At the bottom of the page click on Source Forge to go to the download for the Test Files and download. Extract the files from the .zip file using whichever .zip program you have. Try playing them in NMH. Can you? -

Perhaps you are yet another victim of recent changes that removed the ability to play HD-Audio? There are numerous posts about the issue. Here is an example involving MKV files.
There are a number of free players that you could download and use. I've listed 3 of them here.
For testing, there are some problem files that can be downloaded here. -

Thanks for your help. There seems to be some differences in mkv files. Test 3 from Source Forge won't play with Windows 10 Films & TV but plays perfectly with 5KPlayer. 5KPlayer will play ant.com mkv files and these are converted correctly with Aimersoft HD converter. Ant.com mkv files lose sound when converted with Nero Recode 14 and don't play with sound with Windows 10 Films & TV. Ant.com mkv files are converted correctly with Win X HD Video Converter (I converted to mp4) and when the converted file is converted back to mkv it plays on Media Home.

What does that have to do with NMH, which is what your question is related to?
The audio for Test 3 is mp3. Nothing to do with the fact that Nero disabled copying/recoding HD-Audio in v2016 for people using Win 10 if they have all the updates.
Are you able to play the mkv files in NMH? -

The point here is that not all mkv files are the same. NMH will play all Tests except Test 4. 5K player will play all files but Test 4 starts juddery, MPC HC 64 bit will not play Test 4 either (installed from Sound Forge and its Combined-Community-Codec-Pack-64bit-2015-10-18) but plays the Ant Video Downloader mkv file which neither NMH nor Windows 10 Films & TV will play.
I think now that NMH lacks a few codecs available on freeware (Next PVR will play the Ant Video Downloader mkv file too) but from my end the problem is fixed. -

.mkv is just a container format. The video and audio formats within it can be anything.
Test 4 is a strange file. I can't play it in NMH nor import it into Nero Video. MediaInfo won't provide the video or audio types. I can play it in VLC Player but there is a delay of about 5 seconds before it starts. VLC Player says it's using Vorbis Audio (vorb) and Theora Video (theo). -
